UE Plasmid Miniprep Kit

The UE Plasmid Miniprep Kit utilizes an improved SDS alkaline lysis method combined with a silica-based DNA preparation membrane for rapid and efficient plasmid DNA purification. The system enables extraction of up to 30 μg of high-purity plasmid DNA from 1–4 mL bacterial cultures with OD600 values between 1.0 and 1.5.

Purified plasmid DNA is suitable for downstream molecular biology applications including DNA sequencing, in vitro transcription and translation, restriction enzyme digestion, bacterial transformation, cloning, and routine plasmid analysis workflows.

Frequently Asked Questions

Why do some cultures produce different plasmid yields?

Plasmid yield is strongly influenced by plasmid copy number. Low-copy-number plasmids may require larger bacterial culture volumes.

Can this kit be used for large plasmids such as BACs?

This kit is not recommended for BAC extraction. Specialized large plasmid extraction kits are recommended for those applications.

Why is my plasmid yield low?

Low yield may result from excessive bacterial cells, incomplete lysis, failure to add ethanol to Buffer W2, or use of old bacterial cultures.

Why is there genomic DNA contamination?

Vigorous mixing during lysis or neutralization can cause genomic DNA contamination. Gentle handling during Buffer S2 and Buffer S3 steps is recommended.

Will the Eluent Buffer affect DNA sequencing?

No. The Eluent Buffer contains 2.5 mM Tris-HCl at pH 8.0 and does not interfere with DNA sequencing performance.

Storage & Stability

  • RNase A should be stored at -20°C for long-term storage.
  • Buffer S1 with RNase A added should be stored at 4°C.
  • All other buffers should be tightly closed and stored at room temperature.
  • Before use, add absolute ethanol to Buffer W2 concentrate according to label instructions.
  • If Buffer S2 forms precipitates, warm at 37°C until dissolved before use.
